Abstract
The Gulf Coast Walleye (Sander vitreus) is a genetically unique Walleye population endemic to the southeastern United States and is increasingly threatened due to habitat loss. The current distribution of this population is poorly understood, especially in the upper Tombigbee River Basin in Mississippi. We developed an environmental DNA (eDNA) sampling method for Gulf Coast Walleye by modifying an existing quantitative PCR (qPCR) assay. Our assay tested highly sensitive for Walleye DNA and presented a minimal risk of cross-amplifying co-occurring nontarget species. We then applied the assay to samples from 26 streams in the upper Tombigbee Basin and positive controls. Suboptimal detection of positive controls suggested that our collection methods did not capture enough DNA to ensure consistent detection. Regardless, we detected potential Walleye presence at one field site in Ponta Creek. We present recommendations for improving the sampling methods in future eDNA surveys.
